The basic molecular biology and pharmacology of the endothelin system is well established and there are clear indications that the endothelins are involved in both the physiological regulation of the cardiovascular system and in the pathophysiology of cardiovascular disease. This text provides a current update of the molecular biology and pharmacology of the endothelins and indicates the therapeutic areas for which the development of inhibitors of endothelin production, or antagonists at endothelin receptors, are likely to prove fruitful.
